When stopped, downshifting and upshifting, the clutch works great. Fine and soft. But sometimes, upshifting from 1 to 2, I feel a different sensation on the lever. I squeeze the clutch and almost reaching the end of the stroke, before engaging gear 2, I notice a strange vibration in the lever. It is a minimal sensation, but something different feels different in the hand. In Sport mode you notice something more, in the rest of the modes it doesn't happen.

Clutch too tight?

Just so you know, that setting is just for finger length and rider comfort.. It brings the lever closer or further from the grip. While this will change engagement area, it really does nothing to cable tension.

You're probably feeling the slipper and gripper clutch ramps doing their thing. I've felt it on occasion on every bike I've ridden with that type of clutch.
 
I had the lever in position 5. I have put it in position 2 and I no have that sensation. Now the cable is less stress
That knob controls only how far the lever is from the grip, cable adjustment is made at the cable underneath the rubber dust cover.
Adjust the cable to leave the clutch unloaded, little freeplay.
Then you can play with the knob for your riding style.
 
Not sure about that, but when shifting from 1st to 2nd, you are going through the neutral detent.
Also, I remembered some advice from an old road racer " shifter to next gear with authority and smooth the clutch like a pro "
